Thymosin alpha 1 is a short peptide first isolated from bovine thymus tissue in the early 1970s during fractionation work aimed at identifying factors that influence T cell development. It belongs to a family of acidic thymic peptides, and the original preparations contained several components that were later separated by chromatography. The compound is now produced synthetically rather than extracted from tissue, which removes batch variability tied to animal sourcing. Researchers describe it as an immunomodulatory peptide because laboratory studies show effects on several cell types of the innate and adaptive immune systems.
The molecule consists of 28 amino acid residues with an acetyl group attached to the N-terminal serine. Its sequence is acidic overall, with several glutamic and aspartic acid residues distributed along the chain and no cysteine, so disulfide bridges do not form. The peptide carries a net negative charge at physiological pH. Because the N-terminus is blocked, the intact molecule resists degradation by many aminopeptidases, which contributes to its stability in biological fluids.
The peptide is generated in cells by cleavage of prothymosin alpha, a larger acidic protein encoded by the PTMA gene. Prothymosin alpha is expressed in many tissues, not only in the thymus, and its functions include nuclear roles in chromatin-related processes. The 28-residue fragment corresponds to the N-terminal portion of that precursor. How the cleavage occurs and how the fragment's concentration is regulated remain open questions; circulating amounts are small and difficult to measure reliably with routine assays.

Storage recommendations center on low temperature, dryness, and protection from repeated freezing and thawing. The intact powder is commonly held at 20 degrees below zero Celsius or colder, while a working solution is divided into single-use aliquots to limit freeze-thaw cycles. Buffered saline or phosphate-buffered saline at neutral pH is frequently used as a diluent. Light sensitivity is not well documented, yet amber vials or foil wrapping are common practice for long-term storage of peptide stocks.
Identity and purity are assessed with a small set of standard peptide methods. Reversed-phase high-performance liquid chromatography separates the main peak from truncated or oxidized species, and its area percentage is the usual purity measure. Mass spectrometry confirms the observed molecular mass against the expected value, while amino acid analysis or peptide mapping checks composition and sequence. Specifications for research-grade material are often stated as 95 percent or higher, though the exact limit depends on the supplier and the intended use.
Laboratory supplies of the peptide usually arrive as a lyophilized powder in sealed vials. The powder is hygroscopic, so a vial should be allowed to reach room temperature before it is opened to prevent condensation on the contents. Weighing and transfer are best performed in a low-humidity environment with clean tools. Once dissolved, the solution should be mixed gently rather than vortexed, because foaming and shear can reduce recovery of the peptide.
Overall evidence quality varies considerably. A large share of published reports come from single centers, rely on surrogate immunological markers, or lack adequate control groups. Systematic reviews have highlighted this heterogeneity as a barrier to pooling results. Open questions include which patients, if any, might benefit, what treatment duration is appropriate, and whether any effect is independent of standard care. The peptide is often described as an immune modulator rather than a therapy for one disease, which complicates confirmatory trial design.
Thymosin alpha 1 was identified in 1977 as a component of thymosin fraction 5, a heterogeneous preparation used in early studies of thymic function. Investigators purified the active material and determined its amino acid sequence, which enabled chemical synthesis. Work in the following decades concentrated on T-cell maturation and immune reconstitution in animals and small human cohorts. Early preparations varied in composition, so results from that period are difficult to compare with studies using defined synthetic peptide.
Clinical research has examined the peptide in chronic hepatitis B and C, as a vaccine adjuvant, and in sepsis and oncology settings. Findings across trials are mixed; some report changes in selected immune markers, while others find no clear clinical benefit. Many studies are small and define outcomes differently, which limits comparison. Regulatory approval is confined to a few countries, and the compound is not an approved drug in the United States or most of Europe.
== Biochemistry == Different forms of life with variable origin processes may have appeared quasi-simultaneously in the early Earth. The other forms may be extinct, having left distinctive fossils through their different biochemistry. Metabolism-like reactions could have occurred naturally in early oceans, before the first organisms evolved. Some of these reactions can produce RNA, and others resemble two essential reaction cascades of metabolism: glycolysis and the pentose phosphate pathway, that provide essential precursors for nucleic acids, amino acids and lipids.

As seen in Fig. 1, the engineered aldehyde tag consists of six amino acids. A set of organisms from all domains of life was chosen and the sequence homology of the sulfatase motif was determined. The sequence used is the best consensus for sequences found in bacteria, archaea, worms and higher vertebrates.

== Primary indexes == In addition to the primary indices listed below, Russell publishes Value and Growth versions of each U.S. index. This divides each index roughly in half, separating companies classified as value stocks from those classified as growth stocks. Companies can appear in both the value and growth versions of an index, though the total number of shares between the value and growth versions will equal the number in the main index. The primary indices are:

== Legislation == Orphan drugs generally follow the same regulatory development path as any other pharmaceutical product, in which testing focuses on pharmacokinetics and pharmacodynamics, dosing, stability, safety and efficacy. However, some statistical burdens are lessened to maintain development momentum. For example, orphan drug regulations generally acknowledge the fact that it may not be possible to test 1,000 patients in a phase III clinical trial if fewer than that number are affected by the disease. Government intervention on behalf of orphan drug development takes several forms:
==== Study techniques ==== Much of what is known about cellular morphology changes and the effects of Rho proteins comes from the creation of a constitutively active mutated form of the protein. Mutation of a key amino acid can alter the conformation of the entire protein, causing it to permanently adopt a conformation that resembles the GTP-bound state. This protein cannot be inactivated normally, through GTP hydrolysis, and is thus "stuck on". When a Rho protein activated in this manner is expressed in 3T3 cells, morphological changes such as contractions and filopodia formation ensue. Because Rho proteins are G-proteins and plasma membrane bound, their location can be easily controlled. In each situation, whether it be wound healing, cytokinesis, or budding, the location of the Rho activation can be imaged and identified. For example, if a circular hole is inflicted in a spherical cell, Cdc42 and other active Rhos are seen in highest concentration around the circumference of the circular injury. One method of maintaining the spatial zones of activation is through anchoring to the actin cytoskeleton, keeping the membrane-bound protein from diffusing away from the region where it is most needed. Another method of maintenance is through the formation of a large complex that is resistant to diffusion and more rigidly bound to the membrane than the Rho itself.

Vance describes his upbringing and family background while growing up in Middletown, Ohio, where his mother and her family had moved after World War II from Breathitt County, Kentucky. Vance states that their Appalachian culture valued traits such as loyalty and love of country despite family violence and verbal abuse. Vance recounts his grandparents' alcoholism as well as his mother's history of drug addictions and failed relationships. Vance's grandparents reconciled and became his guardians. His strict but loving grandmother pushed Vance, who went on to complete undergraduate studies at Ohio State University and earned a Juris Doctor degree from Yale Law School. Vance raises questions about the responsibility of his family and local people for their misfortunes. Vance suggests that hillbilly culture fosters social disintegration and economic insecurity in Appalachia. He cites a personal experience where, while working as a grocery store cashier, he saw welfare recipients with cell phones when he could not afford one. Vance's antipathy toward those who seemed to profit from poor behavior while he struggled is presented as a rationale for Appalachia's political swing from voting Democratic to a strong Republican affiliation. Vance tells stories highlighting the lack of work ethic of the local people, including the story of a man who quit his job after expressing dislike over his work hours, and a co-worker with a pregnant girlfriend who skipped work unexcused.
He argued that wounds should be cleaned and sealed up promptly. This was a radical idea; normally this would have been inviting gas gangrene, but Florey proposed leaving that to the penicillin. His recommendations were acted upon, and the War Office established a training course for pathologists and clinicians at the Royal Herbert Hospital, which made use of film that Florey shot in North Africa. Although he intended that penicillin be used to treat the seriously wounded, there were large numbers of venereal disease cases, against which penicillin was particularly effective, and from a military point of view being able to cure gonorrhea in 48 hours was a breakthrough. The supply situation improved, and 20 million units per day were made available for the Allied invasion of Italy in September. Two out of three gas gangrene casualties now survived.
in the 1980s and spread to the UK and Europe through various related forms (e.g. innovation centres, pépinières d'entreprises, technopoles/science parks). The U.S.-based International Business Innovation Association estimates that there are about 7,000 incubators worldwide. A study funded by the European Commission in 2002 identified around 900 incubation environments in Western Europe. As of October 2006, there were more than 1,400 incubators in North America, up from only 12 in 1980. Her Majesty's Treasury identified around 25 incubation environments in the UK in 1997; by 2005, UKBI identified around 270 incubation environments across the country. In 2005 alone, North American incubation programs assisted more than 27,000 companies that provided employment for more than 100,000 workers and generated annual revenues of $17 billion. Incubation activity has not been limited to developed countries; incubation environments are now being implemented in developing countries and raising interest for financial support from organizations such as UNIDO and the World Bank. Within European Union countries, there are different EU and state funded programs that offer support in form of consulting, mentoring, prototype creation, and other services and co-funding for them.
The ABC domain consists of two domains, the catalytic core domain similar to RecA-like motor ATPases and a smaller, structurally diverse α-helical subdomain that is unique to ABC transporters. The larger domain typically consists of two β-sheets and six α helices, where the catalytic Walker A motif (GXXGXGKS/T where X is any amino acid) or P-loop and Walker B motif (ΦΦΦΦD, of which Φ is a hydrophobic residue) is situated. The helical domain consists of three or four helices and the ABC signature motif, also known as LSGGQ motif, linker peptide or C motif. The ABC domain also has a glutamine residue residing in a flexible loop called Q loop, lid or γ-phosphate switch, that connects the TMD and ABC. The Q loop is presumed to be involved in the interaction of the NBD and TMD, particularly in the coupling of nucleotide hydrolysis to the conformational changes of the TMD during substrate translocation. The H motif or switch region contains a highly conserved histidine residue that is also important in the interaction of the ABC domain with ATP. The name ATP-binding cassette is derived from the diagnostic arrangement of the folds or motifs of this class of proteins upon formation of the ATP sandwich and ATP hydrolysis.
There is a risk of misuse and dependence in both patients and non-medical users of alprazolam; alprazolam's high affinity binding, high potency, and rapid onset increase its abuse potential. The physical dependence and withdrawal syndrome of alprazolam also add to its addictive nature. In the small subgroup of individuals who escalate their doses, there is usually a history of alcohol or other substance use disorders. Despite this, most prescribed alprazolam users do not use their medication recreationally, and the long-term use of benzodiazepines does not generally correlate with the need for dose escalation. However, based on US findings from the Treatment Episode Data Set (TEDS), an annual compilation of patient characteristics in substance abuse treatment facilities in the United States, admissions due to "primary tranquilizer" (including, but not limited to, benzodiazepine-type) drug use increased 79% from 1992 to 2002, suggesting that misuse of benzodiazepines may be on the rise. The Centers for Disease Control and Prevention (CDC) reported an 89 percent increase in emergency room visits nationwide related to nonmedical benzodiazepine use between 2004 and 2008. Alprazolam is one of the most commonly prescribed and misused benzodiazepines in the United States. A large-scale nationwide U.S.

Thymosin fraction 5 is a mixture of many peptides obtained from thymus tissue, while thymosin alpha 1 is a single defined 28-residue molecule. The two names appear together in older literature because the purified peptide was first obtained from that mixture.
